Insights into Autosomal Dominant Polycystic Kidney Disease from Genetic Studies

Summary: Autosomal dominant polycystic kidney disease is mainly caused by mutations in PKD1 and PKD2 genes, but may also be caused by mutations in other endoplasmic reticulum protein biosynthetic pathway genes or somatic mosaicism. Genetic testing aids in diagnosis and prognosis, but screening for PKD1 mutations is challenging, and conventional Sanger sequencing is limited in elucidating the causes of atypical polycystic kidney disease.

Ciliopathies and the Kidney: A Review

Summary: Primary cilia are sensory organelles protruding from the apical surface of most cell types, playing important roles in tissue development and signal transduction. Mutations in ciliary-associated proteins result in a group of diseases, with renal ciliopathies being a main focus characterized by kidney cysts due to dysregulated ciliary-dependent signaling. Cystic-associated kidney damage and systemic inflammation lead to kidney failure in these diseases, with no curative pharmacologic treatments currently available.

A Multitubular Kidney-on-Chip to Decipher Pathophysiological Mechanisms in Renal Cystic Diseases

Summary: ADPKD is a major renal pathology caused by deletion of PKD1 or PKD2 genes, leading to local renal tubule dilation, cyst formation, and renal failure in adulthood. A kidney-on-chip model was developed to study the role of adjacent tubule coupling in ADPKD, revealing tube dilation and increased cell proliferation in ADPKD cellular models. The device demonstrated the interplay between tightly packed renal tubes and is a promising tool for further study of kidney pathophysiology.
