Integrated analysis of single-cell RNA-seq and bulk RNA-seq unravels tumour heterogeneity plus M2-like tumour-associated macrophage infiltration and aggressiveness in TNBC

Xuanwen Bao et al.

Summary: Triple-negative breast cancer is characterized by aggressive clinical behavior and significant tumor heterogeneity. Utilizing single-cell and bulk tissue transcriptome analysis can reveal the immune landscape association and predict prognosis and treatment response. In TNBC, a subset of cells with high levels of EMT, stemness, and angiogenesis was identified, while M2-like TAMs were found to be associated with poor prognosis and immunosuppressive characteristics. Machine learning methods and a neural network-based deep learning framework based on TAM-related gene signature show promise in predicting immunotherapy response.

Unraveling novel survivin mRNA transcripts in cancer cells using an in-house developed targeted high-throughput sequencing approach

Panagiotis G. Adamopoulos et al.

Summary: Survivin, a member of the inhibitor of apoptosis protein family, is upregulated in malignant tumors and exists in multiple alternative splicing variants. A novel targeted DNA-seq assay identified fourteen new survivin alternative splice variants, shedding light on their expression profiles and potential biological functions. Further studies will be crucial in understanding how survivin isoforms regulate cell proliferation and apoptosis in cancer, offering new diagnostic and therapeutic opportunities.

Fatigue in inflammatory rheumatic diseases: current knowledge and areas for future research

Kristen Davies et al.

Summary: Fatigue is a significant health concern for individuals with chronic inflammatory rheumatic diseases, but its underlying mechanisms remain unclear. Research suggests roles for the immune system, nervous systems, and psychosocial factors in fatigue, but the interrelationships between these mechanisms are still poorly defined. Further study is needed to better understand and address fatigue in this population.

Membranous nephropathy

Pierre Ronco et al.

Summary: Membranous nephropathy (MN) is a glomerular disease that can occur at any age, with about 80% of adult patients having no apparent cause. It is an autoimmune disease with unpredictable disease outcome, with some patients experiencing spontaneous remission. Various treatment options are available, but relapses and suboptimal treatment responses remain a challenge.

circPTPN22 as a novel biomarker and ceRNA in peripheral blood mononuclear cells of rheumatoid arthritis

Zhuyan Jiang et al.

Summary: High-throughput RNA sequencing revealed differential expression of circRNAs in RA patients compared to HCs, with circPTPN22 identified as a common differentially expressed gene in RA and SLE. The expression of circPTPN22 demonstrated high diagnostic value in distinguishing RA patients from HCs and SLE patients. Furthermore, circPTPN22 levels were positively correlated with various RA-related factors in PBMCs.

Role of NLRP3 Inflammasome in Lupus Nephritis and Therapeutic Targeting by Phytochemicals

Dantong Wu et al.

Summary: Systemic lupus erythematosus (SLE) is a multisystem autoimmune inflammatory condition with severe clinical manifestations. Lupus nephritis (LN) is a major manifestation of SLE, caused by immune complex deposition in the glomeruli. The NLRP3 inflammasome plays a key role in innate immunity and has been implicated in the pathogenesis of autoimmune diseases, including LN.

Novel Long Non-coding RNA Expression Profile of Peripheral Blood Mononuclear Cells Reveals Potential Biomarkers and Regulatory Mechanisms in Systemic Lupus Erythematosus

Qi Cheng et al.

Summary: Transcriptome sequencing identified multiple differentially expressed lncRNAs and mRNAs, with lncRNA NONHSAT101022.2 significantly downregulated in SLE patients and correlated with disease activity and severity. Functional and regulatory analysis suggested that NONHSAT101022.2 may enhance beta 2-AR signal transduction by cis regulating LMBRD2, inducing NK cells to produce high levels of IFN-gamma, thus exacerbating SLE.
