Summary: Wild-type human glutathione peroxidase 4 (GPX4) was efficiently produced in human HEK cells with the co-expression of SBP2, leading to the determination of its crystal structure at 1.0 angstrom resolution. A mass-spectrometry-based approach was developed to monitor the reaction of the active-site selenocysteine with covalent inhibitors, enabling the determination of the crystal structure of GPX4 in complex with the covalent inhibitor ML162 [(S)-enantiomer]. This approach opens the possibility for further crystal structures of GPX4 in complex with potential cancer drug targets.
