Visible-Light-Induced Palladium-Catalyzed Selective Defluoroarylation of Trifluoromethylarenes with Arylboronic Acids

Summary: Selective functionalization of inactive C(sp(3))-F bonds to prepare medicinally interesting aryldifluoromethylated compounds remains challenging. One promising route is the transition-metal-catalyzed cross-coupling through oxidative addition of the C(sp(3))-F bond in trifluoromethylarenes (ArCF3), which are ideal precursors for this process due to their ready availability and low cost. Here, we report an unprecedented excited-state palladium catalysis strategy for selective defluoroarylation of trifluoromethylarenes with arylboronic acids. This visible-light-induced palladium-catalyzed cross-coupling proceeds under mild reaction conditions and allows transformation of a variety of arylboronic acids and ArCF3. Preliminary mechanistic studies reveal that the oxidative addition of the C(sp(3))-F bond in ArCF3 to excited-state palladium(0) via a single electron transfer pathway is responsible for the C(sp(3))-F bond activation.

C-F bond activation under transition-metal-free conditions

While advances have been made in functionalizing activated C-F bonds using transition metal complexes, fewer approaches are available for nonactivated C-F bonds due to the difficulty in oxidative addition of transition metals. Using Lewis acid and light/radical initiator to activate inert C-F bonds has emerged as a powerful tool, providing greener, more economical processes without heavy metal residues in the pharmaceutical industry.

Photo-induced thiolate catalytic activation of inert Caryl-hetero bonds for radical borylation

A novel photocatalytic strategy using thiolate as a catalyst has been introduced for the activation of inert substituted arenes for aryl borylation, expanding the scope of aryl radical precursors and demonstrating good functional-group tolerance. Spectroscopic investigations suggest the formation of a charge-transfer complex as the key step to activate the substrates.

Photoredox relay-catalyzed gem-difluoroallylation of alkyl iodides
