Loss of Furin in β-Cells Induces an mTORC1-ATF4 Anabolic Pathway That Leads to β-Cell Dysfunction

Summary: FURIN, a proprotein convertase, plays an essential role in glucose homeostasis by activating precursor proteins in the secretory pathway. Knockout of FURIN in β-cells leads to glucose intolerance due to smaller islets with reduced insulin content and abnormal granule morphology. The study suggests a model of mTORC1-ATF4 hyperactivation and impaired lysosomal acidification in β-cells lacking Furin, causing β-cell dysfunction.
