The matrix metalloproteinase inhibitor IPR-179 has antiseizure and antiepileptogenic effects

Diede W. M. Broekaart et al.

Summary: Research has shown that the expression of MMPs and TIMPs is persistently dysregulated in the hippocampus of patients with epilepsy and experimental animals. Treatment with the MMP2/9 inhibitor IPR-179 can reduce seizure severity and improve cognitive behavior in rodent models of epilepsy. It also prevents an MMP2/9-dependent switch-off that normally restrains network excitability, suggesting potential for further investigation in clinical trials.

D-Serine Intervention In The Medial Entorhinal Area Alters TLE-Related Pathology In CA1 Hippocampus Via The Temporoammonic Pathway

Stephen Beesley et al.

Summary: This study demonstrates the significant role of the temporoammonic pathway (TAP) in transferring MEA pathology to the CA1 region independently of the perforant pathway in Temporal Lobe Epilepsy. Intervening in the medial entorhinal area (MEA) can rescue hippocampal CA1 neurons and mitigate neuroinflammation in epileptic animals. The findings suggest that the pathology in CA1 is predominantly driven through the TAP, highlighting specific areas for therapeutic intervention.

Genetic Effects of the Schizophrenia-Related Gene DTNBP1 in Temporal Lobe Epilepsy

Hua Tao et al.

Summary: Recent studies have found a genetic association between the DTNBP1 gene and temporal lobe epilepsy (TLE), with a protective allele identified and a novel mutant allele observed exclusively in TLE patients, potentially promoting disease progression. Further exploration of the role of DTNBP1 in TLE is warranted.

MicroRNAs in the development of resistance to antiseizure drugs and their potential as biomarkers in pharmacoresistant epilepsy

Julia Bohosova et al.

Summary: Despite the development of many new antiseizure drugs in the past decade, a significant percentage of patients remain pharmacoresistant. Predicting therapeutic response and identifying biomarkers for pharmacoresistant patients are crucial tasks, with miRNAs emerging as potential new biomarkers in epilepsy.
