The interplay between Glucocerebrosidase, α-synuclein and lipids in human models of Parkinson's disease

Sonia Sanz Munoz et al.

Summary: Mutations in the GBA gene, encoding GCase, are the highest genetic risk factor for Parkinson’s disease, leading to decreased GCase activity and stability which results in the accumulation of lipid substrates and αS. Changes in αS and lipid levels are correlated with alterations in GCase activity/protein levels. Current strategies aim to revert the changes in GCase activity/protein, αS, and lipid levels in the context of PD.

Spectral characteristics of subthalamic nucleus local field potentials in Parkinson's disease: Phenotype and movement matter

Fabio Godinho et al.

Summary: Parkinson's disease patients exhibit differences in STN-LFP beta-band activity between tremor dominant (TD) and postural instability and gait disorder (PIGD) motor phenotypes, especially during rest and movement states. Movement-induced desynchronization patterns in alpha-beta range vary in TD (10-20 Hz) and PIGD patients (21-28 Hz), leading to improved classification accuracy when considering movement information. These findings suggest that STN-LFP beta-band encodes phenotype-movement dependent information in PD patients.

The development of an implantable deep brain stimulation device with simultaneous chronic electrophysiological recording and stimulation in humans

Abhinav Goyal et al.

Summary: Deep brain stimulation (DBS) is commonly used to treat neurological disorders, but there is a need for a fully-implantable DBS system that can chronically record patient-specific electrophysiological biomarkers to improve therapeutic benefit. The Percept PC is the first FDA-approved device capable of simultaneous recordings and stimulation, with potential applications beyond DBS in understanding normal and abnormal neurophysiology.

Should we start integrating genetic data in decision-making on device-aided therapies in Parkinson disease? A point of view

Philippe A. Salles et al.

Summary: Parkinson's disease is a heterogeneous disorder with genetic risk loci and variants influencing clinical profiles and outcomes of device-aided therapies. Personalized symptomatic treatment should be offered due to unique patient presentations and disease progression. Genetic imprint plays a role in clinical milestones and treatment complications, affecting eligibility for various therapies.

Macular microcirculation characteristics in Parkinson's disease evaluated by OCT-Angiography: a literature review

Evita Evangelia Christou et al.

Summary: The retina may serve as a predictive factor for neurodegenerative disorders, with recent use of Optical Coherence Tomography Angiography (OCT-A) in studying macular microcirculation changes in Parkinson's disease (PD). While preliminary evidence suggests OCT-A could be a useful biomarker for cerebral vasculature in PD, further research is needed to confirm its predictive value in clinical practice.
