Photochemical Internalization Enhanced Vaccination Is Safe, and Gives Promising Cellular Immune Responses to an HPV Peptide-Based Vaccine in a Phase I Clinical Study in Healthy Volunteers

Tone Otterhaug et al.

Summary: PCI technology enhances immune responses by releasing endocytosed antigens via light-induced process. This study demonstrates that PCI vaccination combined with Hiltonol adjuvant is safe at low fimaporfin doses and shows promising immune responses in peptide and protein based vaccination.

Developing New Anti-Tuberculosis Vaccines: Focus on Adjuvants

Ana Rita Franco et al.

Summary: Tuberculosis is a major global health concern, and new vaccines are needed to overcome the limitations of the current BCG vaccine. The World Health Organization considers the development of new TB vaccines to be a top priority, with subunit vaccines being promising candidates that may require adjuvants to enhance their efficacy.

Photochemical internalization (PCI)-mediated activation of CD8 T cells involves antigen uptake and CCR7-mediated transport by migratory dendritic cells to draining lymph nodes

Philipp Schineis et al.

Summary: Antigen cross-presentation to cytotoxic CD8+ T cells is crucial for anti-tumor and anti-viral immune responses. Co-encapsulation of photosensitizers and antigens into microspheres with subsequent photochemical internalization has emerged as a promising strategy. In vivo, dendritic cells take up the microspheres in peripheral tissues and migrate to tissue-draining lymph nodes for CD8+ T cell priming.
