Metabolic profiles of multidrug resistant and extensively drug resistant Mycobacterium tuberculosis unveiled by metabolomics

Summary: Comparative sequence analysis has enabled the annotation of millions of genes from organisms across the evolutionary tree. However, defining the physiological factors that contribute to the pathogenicity of the ecologically unusual pathogen Mycobacterium tuberculosis (Mtb) remains poorly understood. The structure and function of a protein, Rv0812, required for Mtb's optimal in vitro fitness has been reported, revealing a bifunctional enzyme that prioritizes PABA over D-Ala/D-Glu biosynthesis.

Metabolic fluxes for nutritional flexibility of Mycobacterium tuberculosis

Elucidating the Antimycobacterial Mechanism of Action of Decoquinate Derivative RMB041 Using Metabolomics

Summary: Tuberculosis, caused by Mycobacterium tuberculosis, remains a leading cause of death globally due to drug resistance and lack of patient adherence. A decoquinate derivative RMB041 has shown promising antimicrobial activity against Mtb, but its mechanism of action remains unknown. Metabolic analysis suggests that the drug primarily affects fatty acid and amino acid metabolism in Mtb, supporting previous findings of its action on cell wall and DNA metabolism.
