T cell immunity to hepatitis C virus: Lessons for a prophylactic vaccine

Robert Thimme

Summary: HCV-specific T cells are essential in determining the outcome of acute infection, yet often fail leading to viral persistence. Limited knowledge about the immunological determinants of viral clearance, persistence and protective immunity is a major obstacle in developing a prophylactic T cell vaccine.

The Impact of HIV- and ART-Induced Mitochondrial Dysfunction in Cellular Senescence and Aging

Madison Schank et al.

Summary: HIV infection and ART treatment can lead to mitochondrial dysfunction, which contributes to cellular exhaustion, senescence, and apoptosis. The compromised mitochondria result in decreased energy production, oxidative stress, and ultimately cellular dysfunction, impacting the overall health and aging of people living with HIV. Identifying and understanding these mechanisms may help in developing improved therapeutics for treating PLHIV.

A Matter of Life or Death: Productively Infected and Bystander CD4 T Cells in Early HIV Infection

Dechao Cao et al.

Summary: Different mechanisms contribute to the death or survival of CD4 T cells following initial HIV infection, involving programmed cell death pathways and an aberrant DNA damage response. Telomere length is positively correlated with survival in HIV-infected cells, with differences in cell death patterns between productively infected and bystander cells. Targeting viral entry or inhibiting DNA damage response pathways could potentially improve the survival of HIV-infected CD4 T cells.

Immune Activation Induces Telomeric DNA Damage and Promotes Short-Lived Effector T Cell Differentiation in Chronic HCV Infection

Lam Nhat Nguyen et al.

Summary: Chronic HCV infection leads to immune activation in CD4(+) T cells with reduced expression of stem cell-like transcription factor T cell factor 1 and telomeric repeat-binding factor 2 (TRF2). Hyperactivation of phosphoinositide 3-kinase/Akt/mammalian target of rapamycin signaling on T cell receptor stimulation promotes inflammation and cellular damage, while inhibiting Akt signaling and enhancing TRF2 expression may provide therapeutic strategies.
