Vector microbiota manipulation by host antibodies: the forgotten strategy to develop transmission-blocking vaccines

Control measures using acaricides are threatened by resistance in vector populations. Alternative strategies that target the vector microbiota and interfere with pathogen development are encouraged. Understanding the immune functions of host antibodies and disrupting the vector microbiota open up possibilities for novel transmission-blocking vaccines.

Summary: Blood-feeding arthropods harbor a common core microbiota that includes certain enteric bacteria benefitting hosts by upregulating genes for essential nutrients, as shown by comparison with non-blood-feeding insects. The enteric bacteria in blood-sucking vectors expressed significantly more genes encoding essential nutrients and also contained more genes for amino acid synthesis and reactive oxygen species detoxification compared to endosymbionts. In contrast, microbes in non-blood-feeding insects expressed fewer gene families related to nutrient categories.

Summary: The study found that two important mosquito vectors (Aedes aegypti and Anopheles gambiae) possess proteins similar to the three Ixodes scapularis proteins with alpha 1-3 GT activity, suggesting they may synthesize glycoconjugates containing alpha -gal that are transferred to sporozoite surfaces. However, Plasmodium parasites lack proteins with similar activity, indicating that the alpha -gal on sporozoite surfaces may be acquired from mosquito glycoconjugates rather than de novo synthesis by the parasites.

Summary: This study demonstrated that immunization targeting key or non-key taxa of tick microbiota in a vertebrate host model significantly impacts the structure and function of tick microbiota. Ticks fed on mice immunized with Escherichia coli showed reduced abundance of Escherichia-Shigella in their microbiota. Anti-tick microbiota vaccines are proven to be a safe, specific, and easy-to-use tool for manipulation of vector microbiome.
