A guide to investigating immune responses elicited by whole-sporozoite pre-erythrocytic vaccines against malaria

Diana Moita et al.

Summary: Efforts have been made to develop efficient vaccines against malaria, with Whole-sporozoite (Wsp) vaccines showing promise in inducing immune responses against Plasmodium parasites. Various approaches for Wsp PE vaccination are currently under evaluation, with clinical trials focusing on assessing protective efficacy and investigating immune responses.

Efficacy of a low-dose candidate malaria vaccine, R21 in adjuvant Matrix-M, with seasonal administration to children in Burkina Faso: a randomised controlled trial

Mehreen S. Datoo et al.

Summary: This study assessed the safety and efficacy of a new candidate vaccine, R21/MM, in African children aged 5-17 months. The results showed that R21/MM vaccine had a favorable safety profile, high immunogenicity, and promising high-level efficacy in African children.

Mechanisms and targets of Fcγ-receptor mediated immunity to malaria sporozoites

Gaoqian Feng et al.

Summary: Understanding mechanisms of antibody-mediated immunity is crucial for developing highly protective vaccines against malaria. Our study identified key roles of Fc gamma-receptor interactions and phagocytosis in humoral immunity, highlighting specific targets and mechanisms that could maximize vaccine efficacy. Antibodies play critical roles in adaptive immune responses to infectious agents, with Fc gamma-receptors being essential for their activity against Plasmodium falciparum sporozoites.

Reduced risk of placental parasitemia associated with complement fixation on Plasmodium falciparum by antibodies among pregnant women

D. Herbert Opi et al.

Summary: The study identified pregnant women who developed antibodies that effectively promoted complement fixation on placental-binding pRBCs, associated with protection against MiP. These complement-fixing antibodies primarily targeted a specific variant of PfEMP1 called VAR2CSA, and complement enhanced the ability of antibodies to inhibit pRBC binding to CSA for increased protection.

Multi-functional antibody profiling for malaria vaccine development and evaluation

D. Herbert Opi et al.

Summary: It is increasingly clear that multiple antibody effector functions play a role in immunity to malaria. Evaluating vaccine candidates should move beyond individual assays or measuring IgG magnitude alone, and instead incorporate a wider range of assays measuring functional antibody responses. A 3-tier approach to implementing assays to inform vaccine evaluation is proposed.
