Cytotoxic CD8+T cells in cancer and cancer immunotherapy

Hans Raskov et al.

Summary: The functions and interactions between the innate and adaptive immune systems play a crucial role in anticancer immunity. Cytotoxic T cells with CD8 as the most effective effectors in the anticancer immune response form the basis of successful cancer immunotherapies. Advancements in genetically modified or synthetic receptors on cytotoxic T cells are being developed for future cancer treatment. Combinatory regimens may optimize treatment effects and reduce adverse events in cancer therapy.

Tumor suppressor gene mutations correlate with prognosis and immunotherapy benefit in hepatocellular carcinoma

Zaoqu Liu et al.

Summary: The study found that HCC patients with TSG non-mutated tumors are more likely to respond positively to immune checkpoint inhibitors, showing higher levels of immune cell infiltration and immune checkpoint expression, leading to longer overall survival. Therefore, TSG subtypes may serve as promising biomarkers for guiding surveillance protocols and immunotherapeutic decisions in patients with HCC.

Josep M. Llovet et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global health challenge with rising incidence projected to exceed 1 million cases by 2025. Infection by hepatitis B and C viruses remains a major risk factor for HCC, while non-alcoholic steatohepatitis is emerging as a more common risk factor in Western countries. Advances in systemic therapies for HCC, including immunotherapies and targeted therapies, are expected to revolutionize the management of this disease.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Prominent Role of Histone Modifications in the Regulation of Tumor Metastasis

Mariam Markouli et al.

Summary: Tumor aggressiveness and progression rely heavily on the coordinated interaction of genetic and epigenetic mechanisms during metastasis, involving various steps such as EMT, anoikis resistance, intra- and extravasation, and colonization of new tissues. Epigenetic mechanisms like DNA methylation, histone modifications, and non-coding RNAs play crucial roles in regulating gene expression related to the metastatic phenotype. Active and repressive histone post-translational modifications, along with transcription factors, are key in controlling genes associated with tumor progression.

Therapeutic Potential of CUDC-907 (Fimepinostat) for Hepatocarcinoma Treatment Revealed by Tumor Spheroids-Based Drug Screening

Wei Liao et al.

Summary: Cancer is the second leading cause of death globally. Traditional drug screening methods often result in unsuccessful clinical trials due to lack of efficacy. In this study, a novel spheroid cell culture device was used to screen anti-hepatocarcinoma drugs, leading to the discovery of CUDC-907 as a potent anti-HCC agent. Further research showed that CUDC-907 inhibits the PI3K/AKT/mTOR pathway and downregulates c-Myc expression, thereby suppressing HCC cells.

Clinical Significance and Inflammatory Landscape of a Novel Recurrence-Associated Immune Signature in Stage II/III Colorectal Cancer

Zaoqu Liu et al.

Summary: The study developed a novel recurrence-associated immune signature (RAIS) to identify high-risk recurrence patients with stage II/III colorectal cancer. The high-risk group showed a higher mutation rate of TP53, while the low-risk group had a more abundant presence of activated CD4+/CD8+ T cells and higher expression of PD-1/PD-L1. The RAIS model was proven to be highly predictive of recurrence and could potentially optimize decision-making in adjuvant chemotherapy and immunotherapy, as well as tailor surveillance protocol for individual patients.
