High tumor mutation burden fails to predict immune checkpoint blockade response across all cancer types

D. J. McGrail et al.

Summary: Analysis of over 10,000 patient tumors from The Cancer Genome Atlas showed that TMB-H tumors had significantly higher response rates to immune checkpoint blockade in certain cancer types, while showing no significant benefit in others. These results do not support the use of TMB-H as a biomarker for ICB treatment across all solid cancer types, indicating the need for further tumor type-specific studies.

Wedding of Molecular Alterations and Immune Checkpoint Blockade: Genomics as a Matchmaker

Elena Fountzilas et al.

Summary: The development of checkpoint blockade immunotherapy has had a positive impact on clinical outcomes, but some patients show resistance and experience serious immune-related adverse events. Understanding the molecular mechanisms and identifying the drivers of therapeutic response and resistance is crucial in optimizing precision immunotherapy.

Phenotypic Characteristics of the Tumour Microenvironment in Primary and Secondary Hepatocellular Carcinoma

Petros Fessas et al.

Summary: This study investigated the intra-tumoural heterogeneity and immune microenvironment of hepatocellular carcinoma (HCC) across primary and secondary sites, revealing that HCC metastases maintain immune exclusion characteristics and exhibit differential regulation of innate immune pathways.

Resistance to Durvalumab and Durvalumab plus Tremelimumab Is Associated with Functional STK11 Mutations in Patients with Non-Small Cell Lung Cancer and Is Reversed by STAT3 Knockdown

Nabendu Pore et al.

Summary: Mutations in the STK11 gene can regulate resistance to PD-1/PD-L1 blockade in patients with nonsquamous non-small cell lung cancer. These mutations may lead to resistance to checkpoint blockade by affecting immune phenotype and immune checkpoints. Modulation of STAT3 signaling can reverse this resistance, suggesting potential combination therapies for STK11-mutant NSCLC.

Dectin-1 Controls TSLP-Induced Th2 Response by Regulating STAT3, STAT6, and p50-RelB Activities in Dendritic Cells

Chao Gu et al.

Summary: The study revealed that Dectin-1 can effectively control TSLP-induced Th2-type immune responses by reducing the expression of OX40L and CCL17. Experimental results showed that Dectin-1 activation can suppress allergic reactions and allergen-specific immune responses in both ex vivo and in vivo settings.

Biomarkers and Gene Signatures to Predict Durable Response to Pembrolizumab in Non-Small Cell Lung Cancer

Anello Marcello Poma et al.

Summary: This study aimed to identify predictive biomarkers for first-line pembrolizumab treatment in NSCLC patients. Patients with durable clinical benefit showed greater infiltration of immune cells, with CD8 T-cell and NK cell scores strongly associated with durable benefit. XCL1/2 demonstrated high performance in predicting durable response to pembrolizumab, outperforming PD-L1 expression levels. These predictive biomarkers can improve patient selection for optimized treatment outcomes.
