Nuclear localization of Zika virus NS5 contributes to suppression of type I interferon production and response

Summary: The study identified NLS-containing regions in ZIKV NS5 that are critical for its nuclear localization. Further experiments showed that the nuclear localization of ZIKV NS5 mainly participates in suppressing IRF3-mediated IFN-I transcription activation and inhibiting IFN-I downstream response, independent of its effect on STAT2 degradation. These findings suggest that NS5's subcellular localization is important for its function in immune suppression, providing new insights into ZIKV pathogenesis.

This study found that human astrocytes infected with ZIKV underwent necroptosis, as evidenced by increased phosphorylation of RIPK1, RIPK3, and MLKL proteins. Inhibition of RIPK3, but not RIPK1, suppressed cell death, indicating RIPK1-independence in ZIKV-induced necroptosis. The findings suggest that necroptosis may play a protective role against viral replication in astrocytes.

Summary: Advances in proteomics have provided a comprehensive understanding of host-pathogen interactions, with a specific focus on Japanese encephalitis virus (JEV) infection-driven changes in mouse embryonic fibroblast (MEF) proteomes. The study identified alterations in innate immune sensing, interferon responses, inflammation, and other biological pathways in response to JEV infection. Functional validation demonstrated the crucial role of anti-viral innate immune proteins in restricting viral replication and the impact of TLR2 signaling in modulating inflammatory cytokine production. Additionally, the study revealed significant downregulation of proteins related to cell adhesion, transport, and lipid biosynthesis in infected cells.
