Early Life Stress Exacerbates Outcome after Traumatic Brain Injury

Chantal M. Sanchez et al.

Summary: Early life stress may contribute to persistent cognitive deficits in individuals with mild traumatic brain injury, including impaired learning and memory function, as well as worsened cortical pathology.

20-HETE synthesis inhibition attenuates traumatic brain injury-induced mitochondrial dysfunction and neuronal apoptosis via the SIRT1/PGC-1α pathway: A translational study

Wenxing Cui et al.

Summary: The study found that the inhibition of 20-HETE synthesis reduced neuronal apoptosis, reactive oxygen species production, and damage to mitochondrial structures after traumatic brain injury (TBI). Mechanistically, the inhibitor also regulated mitochondrial dynamics and promoted the restoration of SIRT1 and PGC-1 alpha, which are crucial in cell function and survival. Moreover, plasma 20-HETE levels were correlated with the neurological outcomes in TBI patients.

Chondroitin 6-sulphate is required for neuroplasticity and memory in ageing

Sujeong Yang et al.

Summary: Perineuronal nets (PNNs), containing chondroitin sulphate proteoglycans on neuronal surfaces, play a role in neuroplasticity and memory. Age-related reduction of chondroitin 6-sulphates (C6S) in PNNs leads to increased inhibition. Manipulating CS composition of PNNs can restore neuroplasticity and memory deficits in aged mice, with a focus on the importance of C6S in memory and neuroplasticity.

Ankyrin-R regulates fast-spiking interneuron excitability through perineuronal nets and Kv3.1b K+ channels

Sharon R. Stevens et al.

Summary: AnkR is highly enriched in Pv(+) fast-spiking interneurons in mice and humans, and is associated with cytoskeletal proteins, cell adhesion molecules (CAMs), and perineuronal nets (PNNs). Loss of AnkR leads to disruption of PNNs, decreases in anxiety-like behaviors, and changes in the excitability and firing properties of Pv(+) fast-spiking interneurons, which is accompanied by a reduction in Kv3.1b K+ channels. AnkR plays a crucial role in organizing ion channels, CAMs, and PNNs in interneurons and at nodes of Ranvier.
