Trained Immunity and Reactivity of Macrophages and Endothelial Cells

Charles Drummer et al.

Summary: Innate immune cells can develop trained immunity after exposure to stimuli like lipopolysaccharides and oxLDL, leading to altered responses upon subsequent challenges. This phenomenon is important for host defense and inflammation, particularly in cardiovascular and metabolic diseases. Trained immunity involves rewiring cellular metabolism and epigenetic remodeling that enhance gene transcription and proinflammatory immune responses, contributing to chronic inflammation.

A Novel Subset of CD95+ Pro-Inflammatory Macrophages Overcome miR155 Deficiency and May Serve as a Switch From Metabolically Healthy Obesity to Metabolically Unhealthy Obesity

Candice Johnson et al.

Summary: Metabolically healthy obesity (MHO) accounts for 35% of obese patients, but there is uncertainty on whether it is a stable condition or a transition to metabolically unhealthy obesity (MUO). This study suggests that MHO may transition to MUO via inflammation driven by CD95(+) pro-inflammatory macrophages.

Trained Immunity: Reprogramming Innate Immunity in Health and Disease

Summary: Trained immunity is a newly recognized phenomenon where cells of the innate immune system exhibit memory characteristics and enhanced responses upon secondary challenge. It explains the heterologous effects of vaccines, but can also lead to maladaptive effects in chronic inflammatory conditions.

Caspase-11-Gasdermin D-Mediated Pyroptosis Is Involved in the Pathogenesis of Atherosclerosis

Mengqing Jiang et al.

Summary: The study shows that oxidized phospholipids can induce pyroptosis via noncanonical inflammasome activation, promoting the development of atherosclerosis. Further experiments demonstrate that inflammatory signaling mediated by caspase-11 and gasdermin D is significantly activated in peripheral blood mononuclear cells from patients with coronary heart disease.

Caspase-11-Mediated Hepatocytic Pyroptosis Promotes the Progression of Nonalcoholic Steatohepatitis

Yingwei Zhu et al.

Summary: The study found that caspase-11 was upregulated in the livers of NASH mice. Caspase-11-deficient mice treated with MCD showed significantly reduced liver injury, fibrosis, and inflammation, as well as inhibited activation of gasdermin D and interleukin-1 beta. Overexpression of caspase-11 promoted steatohepatitis, indicating that caspase-11-mediated hepatocytic pyroptosis promotes the progression of NASH.

Role of the inflammasome, gasdermin D, and pyroptosis in non-alcoholic fatty liver disease

Itzayana Rodriguez-Antonio et al.

Summary: Pyroptosis is a type of programmed cell death mediated by the inflammasome through the pro-inflammatory activity of gasdermin D. Research indicates that gasdermin D plays a crucial role in connecting an elevated immune response to a sustained inflammation, linking it to the progression of non-alcoholic fatty liver disease to non-alcoholic steatohepatitis. Both basic and clinical studies show evidence supporting gasdermin D as a potential therapeutic target for diseases characterized by perpetuated inflammatory processes.

Endothelial Immunity Trained by Coronavirus Infections, DAMP Stimulations and Regulated by Anti-Oxidant NRF2 May Contribute to Inflammations, Myelopoiesis, COVID-19 Cytokine Storms and Thromboembolism

Ying Shao et al.

Summary: Transcriptomic changes in endothelial cells (ECs) infected by coronaviruses and stimulated by DAMPs lead to upregulation of innate immune regulatomic genes (IGs), contributing to various aspects including COVID-19, cardiovascular diseases, inflammation, transplantation, autoimmune diseases, and cancers. The activation of nucleic acid sensors and modulation of gene expression by various PAMPs and DAMPs play crucial roles in the immune response of ECs. The findings suggest a novel function of ECs in promoting myelopoiesis and trained immunity, which may drive cytokine storms and increase thromboembolic potential in COVID-19.
