Role of Basal ABA in Plant Growth and Development

Sensor histidine kinases mediate ABA and osmostress signaling in the moss Physcomitrium patens

Summary: To survive fluctuating water availability, terrestrial plants need to sense water stresses and respond accordingly. The plant hormone ABA and the SnRK2 protein kinase play crucial roles in osmostress responses. In the moss Physcomitrium patens, it was discovered that ABA and osmostress-dependent SnRK2 activation require phosphorylation by an upstream kinase called ARK. Surprisingly, ARK is also involved in regulating the submergence response in P. patens through its ortholog CTR1. However, the activation mechanism of the ARK/SnRK2 module in response to water stress is still unknown. This study found that a group of ethylene-receptor-related sensor histidine kinases (ETR-HKs) is essential for ABA and osmostress responses in P. patens, and the intracellular kinase domain of an ETR-HK physically interacts with ARK. The HK disruptants fail to activate SnRK2 in response to ABA and osmostress due to the lack of ABA-dependent autophosphorylation of ARK's critical serine residue. These findings suggest that the HK/ARK module functions as an integration unit for water availability responses in P. patens.
