Micro RNA Targets in HIV Latency: Insights into Novel Layers of Latency Control

Ashley I. Heinson et al.

Summary: Despite significant progress in identifying cellular factors and pathways contributing to the latent HIV reservoir, the major obstacle to eradicating the virus remains. Noncoding genes, including microRNAs, have been implicated in regulating HIV expression during latency. This study revealed dysregulation of miRNAs in a primary CD4(+) T cell model of HIV latency, with a subset targeting pathways relevant to HIV expression and establishment of latency, such as the p53 signaling pathway.

Cobomarsen, an Oligonucleotide Inhibitor of miR-155, Slows DLBCL Tumor Cell Growth In Vitro and In Vivo

Eleni Anastasiadou et al.

Summary: miRNA-155, an oncogenic miRNA highly expressed in B-cell malignancies, particularly in ABC-DLBCL, represents a potential diagnostic and prognostic biomarker. Inhibition of miR-155 with cobomarsen showed efficacy in reducing cell proliferation and inducing apoptosis in cell lines and xenograft models, with therapeutic application in patients demonstrating reduced and stabilized tumor growth without toxic effects.

miR-150-mediated increase in glucose uptake in HIV-infected cells

Ravi C. Dubey et al.

Summary: The microRNA miR-150 plays a crucial role in HIV infection, increasing viral spread within cells by regulating the expression of glucose transporter-1 (GLUT1). The study reveals that HIV Tat protein enhances miR-150 expression, leading to elevated glucose uptake and promoting viral replication.

Mitochondrial stress induced by continuous stimulation under hypoxia rapidly drives T cell exhaustion

Nicole E. Scharping et al.

Summary: Continuous TCR signaling and hypoxia increase Blimp-1, suppressing mitochondrial reprogramming and generating reactive oxygen species, leading to T cell exhaustion. Mitigating T cell-intrinsic ROS and reducing tumor hypoxia can limit T cell exhaustion, synergizing with immunotherapy.

Transcriptome-wide analysis reveals insight into tumor suppressor functions of 1B3, a novel synthetic miR-193a-3p mimic

Marion T. J. van den Bosch et al.

Summary: miR-193a-3p acts as a suppressive role in many cancers, while its mimic 1B3 demonstrates anti-cancer effects in various human tumor cell lines. The mechanism of action of 1B3 mainly involves upregulation of the PTEN pathway, downregulation of oncogenic growth factor signaling pathways, and modulation of DE genes to exert its anti-cancer effects.

Oncogene-regulated release of extracellular vesicles

Seda Kilinc et al.

Summary: Specific oncogenes reduce cancer cell biomass by promoting extracellular vesicle release, altering protein composition and secreted miRNAs. MYC and AURKB are the most active oncogenes in this process, each requiring different mechanisms for high levels of EV release. This new oncogene-mediated biomass regulation through differential EV release is a novel metabolic phenotype.

Summary: The study revealed that specific metabolites, including guanosine diphosphate (GDP) and uridine diphosphate (UDP) enriched in the TIF of melanoma, can modulate the activity of immune cells and influence the tumor microenvironment. Local administration of UDP in B16 melanoma-engrafted mice showed inhibition of tumor growth and enhancement of anti-tumor immune response, indicating the potential of UDP as an adjuvant in cancer immunotherapy.
