Mitochondrial Dysfunction and Mitophagy in Parkinson's Disease: From Mechanism to Therapy

Ana Belen Malpartida et al.

Summary: Mitochondrial dysfunction has long been associated with Parkinson's disease, although its exact role remains unclear. Recent discoveries on the role of PD familial genes in mediating mitophagy and the overlap with sporadic PD have opened up new opportunities for therapeutically targeting mitochondria in PD. Progress has also been made in understanding the upstream and downstream regulators of canonical and noncanonical PINK1/parkin-mediated mitophagy in response to mitochondrial damage.

Genome sequencing analysis identifies new loci associated with Lewy body dementia and provides insights into its genetic architecture

Ruth Chia et al.

Summary: By conducting whole-genome sequencing and genetic analysis, researchers have identified common genetic risk factors and pathways shared between Lewy body dementia, Alzheimer's disease, and Parkinson's disease. This provides a deeper understanding of the complex genetic architecture of these age-related neurodegenerative conditions.

Pivotal Role of Fyn Kinase in Parkinson's Disease and Levodopa-Induced Dyskinesia: a Novel Therapeutic Target?

Efthalia Angelopoulou et al.

Summary: The exact etiology of Parkinson's disease (PD) is still unknown, and there is no cure for PD. Fyn, a tyrosine phospho-transferase, may play a crucial role in the pathogenesis of PD and levodopa-induced dyskinesia (LID) by regulating key signaling pathways. Therapeutic targeting of Fyn or Fyn-related pathways could represent a novel approach in PD treatment.

Genome-Wide Association Studies of Cognitive and Motor Progression in Parkinson's Disease

Manuela M. X. Tan et al.

Summary: The study found that genetic variants associated with Parkinson's disease risk are not linked to disease progression. The APOE epsilon 4 tagging variant was significantly associated with cognitive progression, while ATP8B2 gene showed a nominal association with motor progression. The new method shows promise in improving the measurement of symptom progression in PD.

Macular microcirculation characteristics in Parkinson's disease evaluated by OCT-Angiography: a literature review

Evita Evangelia Christou et al.

Summary: The retina may serve as a predictive factor for neurodegenerative disorders, with recent use of Optical Coherence Tomography Angiography (OCT-A) in studying macular microcirculation changes in Parkinson's disease (PD). While preliminary evidence suggests OCT-A could be a useful biomarker for cerebral vasculature in PD, further research is needed to confirm its predictive value in clinical practice.
