The RyR2-R2474S Mutation Sensitizes Cardiomyocytes and Hearts to Catecholaminergic Stress-Induced Oxidation of the Mitochondrial Glutathione Pool

Summary: The study demonstrates that the activation of CaMKII delta through Met281/282 oxidation is not necessary for the recovery of Ca2+ transients during acidosis or for the occurrence of postacidic cellular arrhythmias. Despite an usually prooxidant increase in cytosolic Na+, acidosis reduces the cytosolic glutathione redox state within cardiac myocytes, suggesting that oxidation of cytosolic proteins is less likely to occur during acidosis.

Calcium influx through the mitochondrial calcium uniporter holocomplex, MCUcx

Summary: The study quantitatively measured Ca2+ flux into the mitochondrial matrix through MCUcx in heart and skeletal muscle for the first time, and discussed the role of Ca2+ in regulating ATP production in the matrix. The properties and functions of MCUcx subunits, as well as ongoing investigations into the roles of MCUcx in cardiac function, were also highlighted in the review. Additionally, the review introduced new advanced methods for conducting quantitative measurements of Ca2+ in mitochondria while controlling the inner mitochondrial membrane potential.

cAMP Imaging at Ryanodine Receptors Reveals β2-Adrenoceptor Driven Arrhythmias

Summary: This study established a real-time live-cell imaging approach to visualize cAMP levels in the vicinity of cardiac RyR2 in mouse and human failing cardiomyocytes under beta-AR stimulation. The findings revealed a subcellular redistribution of PDEs in cardiac hypertrophy, along with increased beta(2)-AR-dependent RyR2 phosphorylation, sarcoplasmic reticulum calcium leak, and arrhythmia susceptibility in failing mouse and human myocytes. This imaging approach may uncover potential antiarrhythmic targets for heart failure.

This increased propensity for Ca2+ alternans is explained by prolongation of Ca2+ release refractoriness, although it does not surpass that of wild type.

Studies have shown that in CPVT models, cardiomyocytes are more likely to develop SCR upon adrenergic stimulation, potentially resulting in arrhythmias at the tissue level.
