Nuclear localization of Zika virus NS5 contributes to suppression of type I interferon production and response

Zikai Zhao et al.

Summary: The study identified NLS-containing regions in ZIKV NS5 that are critical for its nuclear localization. Further experiments showed that the nuclear localization of ZIKV NS5 mainly participates in suppressing IRF3-mediated IFN-I transcription activation and inhibiting IFN-I downstream response, independent of its effect on STAT2 degradation. These findings suggest that NS5's subcellular localization is important for its function in immune suppression, providing new insights into ZIKV pathogenesis.

The Fatty Acid Lipid Metabolism Nexus in COVID-19

Jerome E. Tanner et al.

Summary: Enteric symptomology in early stages of SARS-2003 and COVID-19 indicates virus replication in the intestine, liver, and pancreas. Aberrant lipid metabolism in morbidly obese individuals worsens the immune response to COVID-19. Targeting lipid metabolism could inhibit virus replication and improve patient response to the disease.

Targeting AMPK by Statins: A Potential Therapeutic Approach

Sajad Dehnavi et al.

Summary: Statins, a group of lipid-lowering drugs, have been shown to regulate multiple proteins associated with different cellular pathways, including the 5'-adenosine monophosphate-activated protein kinase (AMPK) pathway, which plays a significant role in maintaining metabolic homeostasis. Statins can affect the AMPK pathway in various physiological and pathological ways, resulting in therapeutic effects on cancer, cardiovascular diseases, neurodegenerative disorders, and other conditions.

Akt Kinase Intervenes in Flavivirus Replication by Interacting with Viral Protein NS5

Laura Albentosa-Gonzalez et al.

Summary: This study found that Akt kinase plays an important role in flavivirus replication, with specific differences among different viruses. Treatment with Akt inhibitors reduced titers of USUV and ZIKV in cell culture, but not WNV. Further investigations are needed to explore the PI3K/Akt/mTOR pathway as a potential antiviral target.

Multifaceted Role of AMPK in Viral Infections

Maimoona Shahid Bhutta et al.

Summary: Viral pathogens often manipulate host cell regulatory and signaling pathways to create an optimal environment for their growth and survival. AMPK plays a significant role in infection modulation, with viruses altering AMPK signaling activity to facilitate or inhibit viral replication. Understanding the regulation of AMPK signaling following infection can aid in developing more effective therapeutic strategies against viral infectious diseases, including the COVID-19 pandemic.

The importance of viral and cellular factors on flavivirus entry

Carlos Daniel Cordero-Rivera et al.

Summary: Flaviviruses are arthropod-borne pathogens responsible for significant mortality and morbidity worldwide. The viral entry process involves multiple steps such as attachment, internalization, endosomal escape, and capsid uncoating, providing potential targets for developing broad-spectrum antiviral drugs targeting host factors and cellular pathways.

Sherehan Ibrahim et al.

Summary: Metformin, commonly used in treating type 2 diabetes, has shown potential benefits in reducing mortality from infectious diseases and has been linked to reduced mortality in Covid-19 patients according to observational studies. Its mechanisms of reducing mortality from Covid-19 may involve direct antiviral activity and improvement of host health, but the exact reasons remain unclear.
