Smad3 Promotes Cancer-Associated Fibroblasts Generation via Macrophage-Myofibroblast Transition

Philip Chiu-Tsun Tang et al.

Summary: This study identified a novel role of macrophage-myofibroblast transition (MMT) in cancer, showing that MMT cells can de novo generate cancer-promoting cancer-associated fibroblasts (CAFs). Through fate-mapping, RNA velocity, and pseudotime analysis, a subset of CAFs derived from macrophages was confirmed in the tumor microenvironment. Targeting Smad3, a key regulator in the MMT process, effectively blocked CAF formation and cancer progression in vivo, suggesting MMT as a potential therapeutic target for cancer immunotherapy.

SARS-CoV-2 N Protein Induces Acute Kidney Injury via Smad3-Dependent G1 Cell Cycle Arrest Mechanism

Wenbiao Wang et al.

Summary: COVID-19 can cause severe multi-organ damage with kidneys being one of the major targets. The research shows that SARS-CoV-2 N protein interacts with Smad3 to enhance signaling, leading to tubular epithelial cell death and AKI. Targeting Smad3 may offer a novel therapy for COVID-19-related AKI.

DPP4/CD32b/NF-κB Circuit: A Novel Druggable Target for Inhibiting CRP-Driven Diabetic Nephropathy

Patrick Ming-Kuen Tang et al.

Summary: The study revealed that CRP plays a pathogenic role in diabetic nephropathy and DPP4 may serve as a potential therapeutic target. The use of linagliptin effectively blocks CRP-driven renal injury progression.

Mesenchymal Stem/Stromal Cell-Derived Extracellular Vesicles for Chronic Kidney Disease: Are We There Yet?

Alfonso Eirin et al.

Summary: MSC-derived extracellular vesicles (EVs) have the potential to modulate pathways responsible for renal injury, including inflammation, oxidative stress, apoptosis, fibrosis, and microvascular remodeling, leading to renoprotective effects in experimental studies. However, scarce clinical data and challenges such as standardization of methods for EV isolation and characterization, EV fate, duration of EV effects, and effects of cardiovascular risk factors need to be addressed for successful clinical translation.

TGF-β Signaling: From Tissue Fibrosis to Tumor Microenvironment

Jeff Yat-Fai Chung et al.

Summary: TGF-beta signaling plays crucial roles in inflammatory diseases by regulating immunocytes in tissue fibrosis and accelerating tumor development in cancer. The pleiotropic nature of TGF-beta signaling in generating fibrotic TME, containing CAFs and matrix proteins, remains largely unclear, despite recent studies demonstrating its clinical implications in cancer.
