Syncytiotrophoblast stress in preeclampsia: the convergence point for multiple pathways

Christopher W. G. Redman et al.

Summary: Preeclampsia evolves in 2 stages, with a placental problem generating signals to the mother and causing a range of responses. Early-onset preeclampsia is associated with poor placentation and lasts for several months, resulting in fetal growth restriction. Late-onset preeclampsia is part of a common process in all pregnancies, as the placental function declines with increasing chorionic villous packing. The most relevant feature of preeclampsia is syncytiotrophoblast stress, which can be detected in the maternal circulation. Soluble fms-like tyrosine kinase-1 is the best-known biomarker for preeclampsia.

Stromal Cell-Derived Factor (SDF) 2 and the Endoplasmic Reticulum Stress Response of Trophoblast Cells in Gestational Diabetes Mellitus and In vitro Hyperglycaemic Condition

Aline R. Lorenzon et al.

Summary: The study explored the relationship between SDF2 expression and ER response in placental cells exposed to hyperglycaemia, with findings indicating an increase in SDF2 expression in GDM placentae treated with diet but not with insulin therapy. Furthermore, high glucose reduced SDF2 expression in BeWo cells, while insulin treatment reversed the increase in ER stress factors CHOP and sXBP1 induced by high glucose. These results highlight the importance of insulin in mitigating ER stress in placental cells under hyperglycaemic conditions.

At Preeclampsia Diagnosis, Total Cell-Free DNA Concentration is Elevated and Correlates With Disease Severity

Teodora R. Kolarova et al.

Summary: Total cfDNA levels are significantly higher in preeclampsia patients compared to normal pregnancies, while the placental-derived fraction of cfDNA remains similar. In preeclampsia, higher total cfDNA levels are associated with earlier delivery and higher systolic blood pressure.
