Deubiquitylases in developmental ubiquitin signaling and congenital diseases

Summary: Metazoan development from a one-cell zygote to a fully formed organism requires complex cellular differentiation and communication pathways. To coordinate these processes, embryos frequently encode signaling information with the small protein modifier ubiquitin, which is typically attached to lysine residues within substrates. DUBs, a class of similar to 100 human enzymes, critically regulate ubiquitin signaling and control many essential cellular functions and various aspects of human physiology and development. Recent genetic studies have identified mutations in several DUBs that cause developmental disorders.

Botulinum neurotoxin inhibitor binding dynamics and kinetics relevant for drug design

Summary: A natural product analog, NPP, was found to be an effective inhibitor of botulinum neurotoxin type A, showing mixed-type inhibition for the enzyme activity and forming stable complex with the enzyme. Molecular dynamics simulation confirmed that NPP binds near the active site, interfering with the enzyme-substrate binding and inhibiting its cleavage. This insight into the interaction between NPP and BoNT/A LC will aid in developing better strategies against botulism.
