Transcriptomic Profiling in Mice With CB1 receptor Deletion in Primary Sensory Neurons Suggests New Analgesic Targets for Neuropathic Pain

Low-dose cannabinoid receptor 2 agonist induces microglial activation in a cancer pain-morphine tolerance rat model

Summary: The study aimed to investigate the mechanisms of central immune signaling involved in morphine tolerance inhibition by the CB2 agonist AM1241 in cancer pain treatment. Results showed that AM1241 induced microglial activation and upregulation of IL-1 beta and TNF-alpha in the spinal cord and dorsal root ganglion, while the CB2 antagonist AM630 did not show the same effect. This suggests a potential role of the CB2 receptor pathway in regulating inflammation and pain in cancer patients.

STING controls nociception via type I interferon signalling in sensory neurons

Summary: STING is a critical immune regulator that induces type-I interferons and other cytokines to promote immune-cell-mediated clearance of pathogens and neoplastic cells. STING also plays a role in antitumour immunity and has potential as a target for cancer immunotherapy. Additionally, STING is shown to regulate nociception through IFN-I signaling, leading to pain relief in mice and non-human primates.

Summary: Oxidized phospholipids (OxPL) are highly reactive metabolites in the lipidome, produced acutely in inflamed tissue to induce pain. They excite neurons by activating specific ion channels and increase firing rate of nociceptors under inflammatory conditions. Targeting OxPL with specific drugs or antibodies can control acute inflammatory pain.

Summary: Researchers identified new neuronal clusters induced by spared nerve injury (SNI) through single-cell RNA sequencing on dissociated mouse DRG cells, revealing the transcriptomic changes during the process. This study provides a new molecular mechanism basis for understanding the development of neuropathic pain.

Noncanonical Activity of Endocannabinoids and Their Receptors in Central and Peripheral Synapses

Summary: This review discusses new aspects of endocannabinoid functions including biased agonism of endogenous and synthetic cannabinoids, coupling of CB receptors with various G-proteins and beta-arrestins, as well as noncanonical tonic activity of endocannabinoids. The role of endocannabinoids in neurotransmitter release, short-term and long-term potentiation, and synaptic function is highlighted, with the conclusion that the endocannabinoid system has a wider range of modulating effects than previously recognized. Re-evaluation of its functions considering noncanonical features will lead to a better understanding of its role in normal and pathological nervous system functioning.
