Summary: SGLT2 inhibitor Empagliflozin (EMPA) has been shown to suppress cardiomyocyte autophagic cell death, reduce infarct size, improve cardiac function and survival, by inhibiting Na+/H+ exchanger 1 (NHE1) activity and regulating excessive autophagy. These findings provide new insights for drug development targeting NHE1 and autophagy for ventricular remodeling and heart failure treatment after myocardial infarction in both diabetic and non-diabetic patients.

Summary: This study found that dapagliflozin significantly improved pancreatic injury, reduced pancreatic oxidative stress, ER stress, inflammation, and apoptosis, and restored kidney autophagy in obese rats. The results showed that dapagliflozin had higher efficacy in improving body weight, visceral fat weight, plasma cholesterol level, and pancreatic oxidative stress compared to vildagliptin in the model.

Pathophysiology of diabetic kidney disease: impact of SGLT2 inhibitors

Summary: Diabetic kidney disease is a leading cause of kidney failure globally, with over 50% of individuals entering dialysis or transplant programmes in the USA affected by it. Hyperglycaemia is the primary factor in its development, leading to various pathophysiological disturbances that contribute to progressive glomerular sclerosis and declining renal function. Multiple agents used in combination are likely to be needed to effectively slow the progression of the disease.

These findings suggest that SGLT2 inhibitor therapy may be a potential cardioprotective approach for cardiovascular complications in patients undergoing sunitinib treatment, but clinical trials are needed to confirm these effects.
