Pirfenidone attenuates acetaminophen-induced liver injury via suppressing c-Jun N-terminal kinase phosphorylation

Shigeki Tashiro et al.

Summary: In this study, it was found that Pirfenidone can attenuate APAP-induced liver injury by suppressing TUNEL-positive necrosis through blocking JNK phosphorylation. Simultaneous TUNEL and vital PI staining are useful for distinguishing APAP-induced necrosis from typical oncotic necrosis. Pirfenidone shows potential as a new option for preventing APAP-induced liver injury and holds value for further research and clinical applications.

Inhibition of proliferation-linked signaling cascades with atractylenolide I reduces myofibroblastic phenotype and renal fibrosis

Yangyang Guo et al.

Summary: The study demonstrated that ATL-1 inhibits the myofibroblastic phenotype and fibrosis development in the kidneys by targeting fibroblast-myofibroblast differentiation and epithelial-mesenchymal transition. This anti-fibrotic effect of ATL-1 is associated with the inhibition of proliferation-linked cascades signaling pathways.

IL-22-mediated renal metabolic reprogramming via PFKFB3 to treat kidney injury

Wei Chen et al.

Summary: The study shows that interleukin-22 (IL-22) can attenuate kidney injury by reprogramming the metabolism of renal tubular epithelial cells (TECs) and alleviating mitochondrial reactive oxygen species (ROS) accumulation. IL-22 improves kidney function in cisplatin-induced kidney damage and diabetic nephropathy (DN) animal models by inducing AMPK/AKT signaling and PFBFK3 activities.

Alcohol Abstinence Rescues Hepatic Steatosis and Liver Injury via Improving Metabolic Reprogramming in Chronic Alcohol-Fed Mice

Aiwen Pi et al.

Summary: Alcohol withdrawal effectively restored hepatic lipid metabolism, reversed liver injury and inflammation by improving metabolism reprogramming in the Lieber-DeCarli mouse model of ALD. This study enhances understanding of the biological mechanisms involved in the beneficial role of alcohol abstinence as a treatment for ALD.

Hepatic NF-κB-Inducing Kinase and Inhibitor of NF-κB Kinase Subunit α Promote Liver Oxidative Stress, Ferroptosis, and Liver Injury

Xiao Zhong et al.

Summary: Drug-induced hepatotoxicity poses a challenge to the development of new medications due to the generation of reactive oxygen species during drug metabolism in hepatocytes. This study reveals a previously unrecognized NIK/IKKα/ROS/ferroptosis axis involved in liver disease progression, shedding light on a new mechanism of drug-induced liver injury.
