GPR10 gene deletion in mice increases basal neuronal activity, disturbs insulin sensitivity and alters lipid homeostasis

Veronika Prazlenkova et al.

Summary: Deletion of GPR10 gene resulted in changes in lipid metabolism in mice of both sexes, however in different extent. The increase in adipose tissue mass observed in only GPR10 KO males may have been prevented in GPR10 KO females owing to a compensatory increase in the expression of metabolic genes.

Obesity-induced changes in human islet G protein-coupled receptor expression: Implications for metabolic regulation

Patricio Atanes et al.

Summary: This study compared the expression of GPCR mRNA in human islets obtained from donors with normal weight range and those classified as obese. The altered expression of GPCRs in obesity may have an impact on islet function, and the study also considered the possible effects of adipokines on GPCRs showing altered expression in obesity.

Palmitoylation of Prolactin-Releasing Peptide Increased Affinity for and Activation of the GPR10, NPFF-R2 and NPFF-R1 Receptors: In Vitro Study

Alena Karnosova et al.

Summary: Lipidization of PrRP stabilizes the peptide and increases its affinity for anorexigenic receptors GPR10 and NPFF-R2. Two palmitoylated PrRP31 analogs showed high affinity for NPFF-R1 and increased phosphorylation of ERK, Akt, and CREB in receptor-expressing cells. Palm(11)-PrRP31 exhibited fewer off-target activities and has potential as an anti-obesity drug.

Cholecystokinin System Is Involved in the Anorexigenic Effect of Peripherally Applied Palmitoylated Prolactin-Releasing Peptide in Fasted Mice

Summary: This study suggests that the CCK receptor and PrRP neuronal system play important roles in the central anorexigenic effect. The exogenously influenced CCK system may be involved in the central anorexigenic effect of peripherally applied palm-PrRP31.
