Repurposing of phytomedicine-derived bioactive compounds with promising anti-SARS-CoV-2 potential: Molecular docking, MD simulation and drug-likeness/ADMET studies

In silico analysis of phytochemicals as potential inhibitors of proteases involved in SARS-CoV-2 infection

Summary: In this study, six phytochemicals and three drug compounds were analyzed for their potential as inhibitors against various drug targets of SARS-CoV-2 and human cellular proteins. The results showed that flabelliferin exhibited strong inhibitory effects on viral proteases, while nelfinavir was the most effective inhibitor against COVID-19 main protease. Flabelliferin also showed promising inhibitory effects on human cellular transmembrane serine proteinase. These findings suggest that these compounds could be potential therapeutics for COVID-19, but further clinical studies are required to confirm their efficacy.

Cerebroprotective effect of Aloe Emodin: In silico and in vivo studies

Summary: This study investigated the cerebroprotective potential of aloe emodin (AE) against cerebral injury in rats. Molecular docking analysis was used to evaluate the binding affinity between AE and proteins involved in apoptosis and inflammation. In vivo experiments were conducted to assess the activity of AE on cerebral ischemic reperfusion injury. The results showed that AE exhibited interaction with cerebrotoxic proteins and demonstrated potential inhibition of these proteins. AE also showed anti-inflammatory, antioxidant, and antiapoptotic properties.

In silico screening of phytopolyphenolics for the identification of bioactive compounds as novel protease inhibitors effective against SARS-CoV-2

Summary: This study identified 31 plant-derived polyphenolic compounds with strong binding affinity, of which 6 compounds showed significant binding affinities against SARS-CoV-2 proteases, with taxifolin and morin exhibiting the highest affinities. Molecular dynamics simulation studies indicated structural perturbations of the proteases upon binding, suggesting potential antiviral activities of these compounds against COVID-19. Further in vivo and in vitro studies are warranted to assess their efficacy for the development of potent antiviral agents.

Drug repurposing for coronavirus (COVID-19): in silico screening of known drugs against coronavirus 3CL hydrolase and protease enzymes

Summary: This study attempted to identify commercially available drugs for repurposing against coronavirus through structure-based virtual screening, successfully finding multiple potential inhibitors. The 3D structure of TMPRSS2 was generated using homology modeling, and promising inhibitors were identified through docking studies. ADMET profiles showed that the compounds discovered in the study are safe and drug-like.

Scientists are now exploring potential treatments, including herbal medicines, by studying the structural components and proteins of the virus. Through in silico studies and molecular docking, potential anti-viral properties of certain chemical constituents of herbal medicines are being investigated to target key enzymes of the virus and control viral replication.
