Assessment of pulmonary toxicity of potential antioxidant drug PEGylated nanoceria after intratracheal instillation in rats

Summary: Cerium oxide nanoparticles have unique redox properties and act as excellent antioxidants in biological environments. Research has shown that these nanoparticles could be used as effective antioxidant drugs for various diseases. However, the toxicity of these nanoparticles in living organisms remains controversial and requires further investigation. This study focused on investigating the pulmonary and systemic toxicity of PEGylated CeO(2) nanoparticles in rats and found that high doses induced mild pulmonary inflammation but did not cause severe systemic toxicity.

Nanocatalytic Theranostics with Glutathione Depletion and Enhanced Reactive Oxygen Species Generation for Efficient Cancer Therapy

Summary: Chemodynamic therapy (CDT) is a promising method for killing cancer cells by converting intracellular hydrogen peroxide (H2O2) into highly toxic hydroxyl radicals ((OH)-O-center dot). An intelligent nanocatalytic theranostics, PGC-DOX, was developed to address the limitations of endogenous H2O2 and high GSH levels in tumor cells, leading to efficient cancer therapy. The system combines H2O2 self-supply, GSH-elimination, and DOX-induced chemotherapy to effectively inhibit tumor growth with minimal side effects in vivo.

Reactive Oxygen Species-Regulating Strategies Based on Nanomaterials for Disease Treatment

Summary: ROS play a vital role in physiological and pathological processes, and research on regulating ROS for disease treatments has gained wide attention. Advances in nanotechnology have led to the development of numerous nanomaterials with the ability to regulate ROS, aiming to find new and effective ROS-related nanotherapeutic modalities. Despite significant progress in ROS-based nanomedicines, there is a need to address fundamental principles for designing ROS-associated nanomedicines to minimize gaps in biomedical applications. This review summarizes current progress in ROS-associated nanomedicines in disease treatments and presents key principles for their design. Future perspectives in the development of ROS-associated nanomedicines are also discussed.

Precision engineering the structure and active site towards atomic levels is a trend for nanomedicines, promoting the evolution of metal-based biomedical nanomaterials from nanoparticles to clusters and now to SACs.

High-Performance Self-Cascade Pyrite Nanozymes for Apoptosis-Ferroptosis Synergistic Tumor Therapy

Summary: The pyrite peroxidase nanozyme demonstrates ultra-high affinity for H2O2 and possesses intrinsic glutathione oxidase-like activity, resulting in abundant production of (OH)-O-center dot and depletion of reduced glutathione, inducing apoptosis and ferroptosis of tumor cells. It shows potential for effective therapeutic application in tumor catalytic therapy.

Multi-enzyme mimetic ultrasmall iridium nanozymes as reactive oxygen/ nitrogen species scavengers for acute kidney injury management

Summary: This study introduced ultrasmall polyvinylpyrrolidone-coated iridium nanoparticles as a promising new artificial enzyme to scavenge RONS and protect against AKI. The nanoparticles showed preferential renal uptake and alleviated clinical symptoms in mice, with relatively low systemic side effects due to rapid renal clearance. The research demonstrates the clinically translatable potential of ultrasmall nanozymes for AKI management.

A core-shell Au@Cu2-xSe heterogeneous metal nanocomposite for photoacoustic and computed tomography dual-imaging-guided photothermal boosted chemodynamic therapy

Summary: Chemodynamic therapy (CDT) has gained attention for its specificity in conquering cancers. Through the use of core-shell Au@Cu2-xSe NPs, photothermal conversion efficiency is boosted up to 56.6%, generating heat to accelerate Fenton-like reactions and induce apoptotic cell death. In both in vitro and in vivo studies, tumors were significantly suppressed after this synergistic therapy, guided by PA and CT imaging.

A Titanium Nitride Nanozyme for pH-Responsive and Irradiation-Enhanced Cascade-Catalytic Tumor Therapy

Summary: The study found that nitrogen doping significantly enhanced the peroxidase activity of titanium-based nanozymes, and further improved the catalytic activity of nanozymes under near-infrared laser irradiation. By using TLGp, combining pH-responsive GOx-mediated H2O2 self-supply, nitrogen doping, and irradiation-enhanced enzymatic activity of titanium nitride nanoparticles, along with mild photothermal therapy, effective tumor inhibition with minimal side effects in vivo was achieved.

Summary: The development and metastasis of tumors are closely linked to the tumor microenvironment (TME). Nanoparticles, with their unique physical properties and design, show potential in efficiently penetrating TME and delivering therapeutic agents to specific components. However, the heterogeneity of tumors and individuals may limit the effectiveness of nanoparticle delivery systems in all types of tumors, calling for further understanding of TME changes at different stages of tumor development for the design of more personalized nanoplatforms.

Summary: Polydopamine nanoparticles were utilized to stabilize a nano-Pt catalyst for relieving tumor hypoxia in photodynamic therapy (PDT). In addition to serving as a carrier for nano-Pt and photosensitizers, polydopamine also acted as a photothermal reagent in photothermal therapy (PTT). The combined PDT and PTT mechanism presented an enhanced anti-tumor therapy effect.
