Molecular-based detection, genetic characterization and phylogenetic analysis of porcine circovirus 4 from Korean domestic swine farms

Van-Giap Nguyen et al.

Summary: The novel and unclassified porcine circovirus 4 (PCV4) was first reported in China in 2019 and has been found to be actively circulating in pigs in both China and Korea. The Korean PCV4 strain shares high genetic similarity with other known PCV4 strains. The study also identified key genomic features and predicted essential replication elements for circoviruses in PCV4 strains, contributing to a better understanding of the genetic relationships between PCV4 and other CRESS DNA viruses.

Molecular detection and phylogenetic analysis ofPorcine circovirus4 in Henan and Shanxi Provinces of China

Run-Bo Tian et al.

Summary: A new circovirus, PCV4, was identified in pigs with severe clinical disease in Hunan Province, China in 2019. The virus was detected in samples from pigs with different clinical presentations and showed genetic diversity. The study's results provide new insights into the molecular epidemiology of PCV4.

Development and application of a quadruplex real-time PCR assay for differential detection of porcine circoviruses (PCV1 to PCV4) in Jiangsu province of China from 2016 to 2020

Nanhua Chen et al.

Summary: There are four species of porcine circoviruses (PCVs) and a quadruplex real-time PCR assay has been developed in this study to differentiate PCV1-4 with satisfactory results. The assay showed 100% agreement with conventional PCR assays when applied to clinical samples from Jiangsu province in China, confirming the coexistence of all species of PCV1-4 strains in recent years.

Detection and genetic characterization of porcine circovirus 4 (PCV4) in Guangxi, China

Wenchao Sun et al.

Summary: The study investigated the prevalence and genome diversity of PCV4 in pigs from Guangxi Province, China, between 2015 and 2019. Results showed that PCV4 shared nucleotide similarity with other representative circovirus genomes and was most closely related to bat-associated circovirus and mink circovirus. This study provides insight into the molecular epidemiology of PCV4 in Guangxi Province, China.

Retrospective surveillance of porcine circovirus 4 in pigs in Inner Mongolia, China, from 2016 to 2018

Zhuo Ha et al.

Summary: The study in Inner Mongolia, China, revealed the presence of PCV4 infection in pig farms from 2016 to 2018, with an increasing infection rate over the years. Interestingly, PCV4 infections were not associated with PCV2 or PCV3 infections. Sequencing of three PCV4 strains showed high genetic stability with less than 80% sequence identity to other circoviruses. Phylogenetic analysis confirmed that PCV4 strains formed a distinct branch.

The serine-48 residue of nucleolar phosphoprotein nucleophosmin-1 plays critical role in subcellular localization and interaction with porcine circovirus type 3 capsid protein

Jianwei Zhou et al.

Summary: The research discovered a previously unidentified nucleolar localization signal (NoLS) of PCV3 Cap, which can hijack the nucleolar phosphoprotein NPM1 for nucleolar localization. Both the NoLS of PCV3 Cap and the serine-48 residue of NPM1 are essential for their interaction. The charge property of serine-48 residue of NPM1 plays a critical role in nucleolar localization and interaction with PCV3 Cap.
