Differential expression of microRNAs and tRNA fragments mediate the adaptation of the liver fluke Fasciola gigantica to its intermediate snail and definitive mammalian hosts

Rui-Si Hu et al.

Summary: The study investigated the transcriptional changes of small RNAs during different developmental stages of the tropical liver fluke Fasciola gigantica, revealing important roles of miRNAs and tRFs in parasite development. Furthermore, comparison of miRNA composition between F. gigantica and Fasciola hepatica showed a high level of conservation with some distinct differences, which could be used for species differentiation.

Comprehensive annotation and characterization of planarian tRNA and tRNA-derived fragments (tRFs)

Vairavan Lakshmanan et al.

Summary: tRNA-derived fragments (tRFs) are dynamically expressed during planarian regeneration, suggesting a possible role in regulating regeneration processes. Spatial profiling reveals diverse expression patterns of tRFs, with some enriched in specific regions. 5'-tsRNA interacts with SMEDWI proteins and AGO1 plays a role in processing itRFs, indicating a novel regulatory mechanism in planarian regeneration.

Gianni Frascotti et al.

Summary: The vault nanoparticle is a eukaryotic ribonucleoprotein complex consisting of 78 individual 97 kDa-major vault protein (MVP) molecules that form two symmetrical, cup-shaped, hollow halves. It plays diverse cellular functions including protection against cellular stresses and activation of prosurvival signaling pathways. Its biological roles in physiological and pathological processes may provide new insights for its utilization as a nanocarrier for drug delivery.

Fasciola hepatica hijacks host macrophage miRNA machinery to modulate early innate immune responses

Nham Tran et al.

Summary: The parasite Fasciola hepatica regulates host innate immune responses by releasing miRNA, especially fhe-miR-125b, which mimics host miRNA to negatively regulate the production of inflammatory cytokines. This manipulation of the miRNA machinery controlling innate cell function is a fundamental mechanism by which worm parasites disarm the early immune responses of their host to ensure successful infection.

Expression profiling of Echinococcus multilocularis miRNAs throughout metacestode development in vitro

Natalia Macchiaroli et al.

Summary: The neglected zoonotic disease alveolar echinococcosis (AE) is caused by the metacestode stage of the tapeworm parasite Echinococcus multilocularis. MicroRNAs play important roles in regulating gene expression. The research identified 37 miRNAs in E. multilocularis, some of which are absent in the host. The study also revealed highly regulated miRNAs during different transitions or cultured conditions and showed that miR-71 is expressed in germinative cells and other cell types in E. multilocularis metacestodes.

Extracellular Vesicles: Schistosomal Long-Range Precise Weapon to Manipulate the Immune Response

Dror Avni et al.

Summary: Schistosomiasis, caused by the helminth parasite Schistosoma, affects over 240 million people globally. The parasites release extracellular vesicles (EVs) containing miRNAs that modulate the host immune response, creating a favorable environment for parasitic survival. Understanding the bioactive cargo and selective targeting of these EVs may lead to the development of new tools for diagnostics and therapy for schistosomiasis and immune-related disorders.

The challenging world of extracellular RNAs of helminth parasites

Marcela A. Cucher et al.

Summary: In recent years, cell free or extracellular RNAs (ex-RNAs) have emerged as novel intercellular messengers between animal cells, including pathogens. They have been found to play important roles in infectious diseases and have been studied in parasitic helminths. Protected by encapsulation in extracellular vesicles (EV) or association to proteins or lipoproteins, ex-RNAs have been detected in the excretion/secretion products of helminth parasites.

Summary: Helminth parasites adapt to their complex lifecycles by expressing a unique repertoire of genes at different developmental stages, including the release of microRNAs (miRNAs) and extracellular vesicles (EVs) which aid in host-parasite interactions, immune regulation, and infection processes. These parasite-derived molecules have potential as novel diagnostic tools in the mammalian host circulatory system.

RI-SEC-seq: Comprehensive Profiling of Nonvesicular Extracellular RNAs with Different Stabilities

Juan Tosar et al.

Summary: Exosomes and other extracellular vesicles are important for transporting RNAs, but a significant portion of extracellular RNAs do not co-purify with vesicles, leading to a biased non-vesicular exRNA profile. A new method using size-exclusion chromatography has allowed for the identification and study of extracellular ribosomes and tRNAs, offering a dynamic view of the extracellular RNAome that can impact biomarker discovery.
