microRNAstrand selection: Unwinding the rules

Jeffrey C. Medley et al.

Summary: miRNAs play a central role in regulating gene expression by targeting specific mRNAs for degradation or translational repression. The process of miRNA strand selection determines the specificity of miRISC towards its targets, and aberrant strand selection can lead to altered gene regulation in diseases. Understanding the rules governing miRNA strand selection can shed light on disease mechanisms and potential therapeutic interventions.

Transcriptome-wide analysis reveals spatial correlation between N6-methyladenosine and binding sites of microRNAs and RNA-binding proteins

Sukhen Das Mandal et al.

Summary: m(6)A-modified mRNAs are more targeted by miRNAs and RBPs, with miRNA target sites and RBP-binding sites located close to m(6)A sites, indicating a three-way interplay between m(6)A, microRNA, and RBP binding.

Screening by deep sequencing reveals mediators of microRNA tailing in C. elegans

Karl-Frederic Vieux et al.

Summary: The study identifies that microRNA tailing in Caenorhabditis elegans mainly consists of mono-uridylation of mature microRNA species, and disrupting tailing does not alter microRNA decay. This research paves the way to explore more specialized regulatory roles of tailing in specific contexts or miRNAs.

CSDE1 attenuates microRNA-mediated silencing of PMEPA1 in melanoma

Pavan Kumar Kakumani et al.

Summary: The study reveals that CSDE1 interacts with miRNA-induced silencing complex miRISC in melanoma, affecting the expression of PMEPA1 gene by binding with AGO2. Additionally, miRNA miR-129-5p also represses PMEPA1 expression in melanoma, collectively regulating the impact of PMEPA1 on the progression of melanoma.

Ago2 protects Drosophila siRNAs and microRNAs from target-directed degradation, even in the absence of 2′-O-methylation

Elena R. Kingston et al.

Summary: In Drosophila cells, some siRNAs loaded into different Argonaute paralogs exhibit differential sensitivity to Dora, with those loaded into Ago1 being sensitive to Dora while those loaded into Ago2 being insensitive. This suggests that the protection of siRNAs from Dora-mediated degradation is conferred by features of the Ago2 protein itself rather than 2'-O-methylation at their 3' termini.

The RabGAP TBC-11 controls Argonaute localization for proper microRNA function in C. elegans

Pascale Michaud et al.

Summary: The study demonstrates that TBC-11 plays a crucial role in the miRNA pathway through RAB-6, emphasizing the importance of intracellular trafficking for miRNA function. Proper regulation of the intracellular location of Argonaute protein is essential for ensuring effective microRNA-mediated mRNA repression in animals.

RNA-binding proteins La and HuR cooperatively modulate translation repression of PDCD4 mRNA

Ravi Kumar et al.

Summary: Posttranscriptional regulation of gene expression is crucial in controlling the inflammatory response and preventing tumorigenesis. The interactions between HuR and La proteins play a cooperative role in regulating PDCD4 mRNA translation and mitigating the oncogenic function of miR-21. This interplay demonstrates a synergistic effect on PDCD4 expression, helping maintain balance between inflammation and tumorigenesis.

E-cadherin Interacts With Posttranslationally-Modified AGO2 to Enhance miRISC Activity

Jie-Ning Li et al.

Summary: MicroRNAs (miRNAs) are small non-coding RNAs that play a role in regulating pathophysiological processes by suppressing target gene expression post-transcriptionally. The function of miRNAs is regulated by a conserved biogenesis machinery involving processing steps and the formation of a miRNA-induced silencing complex (miRISC). This study discovered a novel regulatory mechanism involving the post-translational modification of AGO2 by E-cadherin, leading to enhanced miRISC activity on gene silencing.
