Recent Advances in Dissolution Testing and Their Use to Improve In Vitro-In Vivo Correlations in Oral Drug Formulations

Alok Ranjan et al.

Summary: The bioavailability of oral drug formulations is influenced by the composition of gastrointestinal fluids and hydrodynamic conditions, but these factors are difficult to fully characterize in vivo, leading to the use of in vitro experiments. In vitro-in vivo correlation (IVIVC) involves mathematical relationships between in vitro dissolution behavior and in vivo drug concentration in plasma, which can be improved by better mimicking in vivo conditions in vitro. Artificial gastrointestinal systems are designed to help achieve this goal, and ex vivo approaches also play a role in improving IVIVC.

Ethyl Cellulose and Hydroxypropyl Methyl Cellulose Blended Methotrexate-Loaded Transdermal Patches: In Vitro and Ex Vivo

Muhammad Shahid Latif et al.

Summary: Transdermal drug delivery systems (TDDSs) are innovative methods for drug delivery through the skin, aiming to achieve systemic effects. The study focused on designing and optimizing methotrexate-loaded transdermal patches with different concentrations of ethyl cellulose (EC) and hydroxypropyl methyl cellulose (HPMC) for local management of psoriasis. The F5 formulation showed 82.71% sustained drug release and non-Fickian diffusion, with good stability for 180 days.

Chitosan-Coated 5-Fluorouracil Incorporated Emulsions as Transdermal Drug Delivery Matrices

Taif Ali Khan et al.

Summary: The study developed chitosan-coated emulsions containing 5-fluorouracil (5-FU) as a model drug and evaluated their drug release and skin permeation properties. Different surfactants showed varying degrees of skin drug retention, while chitosan-modified emulsions exhibited good drug release control and skin permeation. The emulsions containing a combination of surfactants (Tween) and a co-surfactant (PEG) showed the best penetration profile and potential for transdermal drug delivery.

Ultraflexible Liposome Nanocargo as a Dermal and Transdermal Drug Delivery System

Kalvatala Sudhakar et al.

Summary: The incorporation of active pharmaceutical ingredients into cargo carriers, especially in the form of liposomes, offers elegant solutions for various drug delivery challenges. The transformation of lipids/polymers into vesicular formulations has provided more efficient and professional amalgamation of APIs, ultimately improving bioavailability and solubility. Ultraflexible liposomes have emerged as novel carriers with promising applications in drug delivery systems, overcoming obstacles such as poor solubility and immune detection.

Summary: The study focused on the development, optimization, characterization, and effects of PO-loaded alginate microbeads in terms of drug release, antioxidant, and anti-inflammatory properties. The optimized microbeads showed improved drug release in intestinal fluid and excellent mucoadhesive and swelling properties. In vivo, the microbeads were well-distributed in the intestine and exhibited potential antioxidant effects and anti-inflammatory effects on IBS-induced rats.

Recent advances in transdermal drug delivery systems: a review

Woo Yeup Jeong et al.

Summary: Transdermal drug delivery systems (TDDS) offer a non-invasive alternative to conventional needle injections, with advantages including low rejection rate, convenience, and applicability to pharmaceuticals and cosmetics. Despite obstacles presented by skin physicochemical properties, research has made progress in overcoming these challenges to harness the high efficiency inherent to TDDS.
