Identification of Small-Molecule Inhibitors of Neutral Ceramidase (nCDase) via Target-Based High-Throughput Screening

Over 650,000 small molecules were screened, leading to the identification of several lead series of hits. Researchers have discovered a new series of potent compounds unrelated to praziquantel, with predictions suggesting they could cure infection in humans with a single oral dose 10-fold smaller than praziquantel. Through optimization and design, several compounds in the series have shown efficacy in clearing both juvenile and adult S. mansoni worms in mice with a single oral dose of less than 10 mg/kg, indicating potential for treating schistosomiasis in humans with doses less than 5 mg/kg.
