SORL1 deficiency in human excitatory neurons causes APP-dependent defects in the endolysosome-autophagy network

Summary: The dysfunction of the endolysosomal-autophagy network is considered an important pathogenic process in Alzheimer's disease, and mutations in the SORL1 gene have been found to contribute to this dysfunction. Experimental results show that SORL1 deficiency leads to abnormal endolysosome function in neurons, with complete loss of SORL1 causing additional defects in lysosome function and autophagy. The study demonstrates a common pathway involving SORL1, PSEN1, and APP in regulating the endolysosome system, which becomes dysfunctional in Alzheimer's disease.
