Topoisomerase-Mediated DNA Damage in Neurological Disorders

Gene Size Matters: An Analysis of Gene Length in the Human Genome

Summary: Gene length is strongly positively correlated with transcript length, protein size, genetic variants, and introns in the human genome. Longer gene transcripts are expressed in specific tissues like blood vessels, nerves, thyroid, cervix uteri, and the brain, while shorter transcripts are found in tissues like pancreas, skin, stomach, vagina, and testis. Natural selection suppresses changes for genes with longer transcripts and promotes changes for genes with smaller transcripts. Genes with longer transcripts tend to have more co-expressed genes, protein-protein interactions, and associated publications, while larger transcripts are often linked to neuronal development and smaller transcripts play roles in skin development and the immune system. Additionally, pathways related to cancer, neurons, and heart diseases tend to have genes with longer transcripts, while immune responses and neurodegenerative diseases are associated with genes having smaller transcripts. The study suggests that longer genes play a role in early development stages, while smaller genes are important for functions requiring fast responses throughout life, such as the immune system.

This study provides cryo-EM structures of the entire Topo II alpha nucleoprotein complex, shedding light on the allosteric connections between different domains and the role of the non-conserved C-terminal domain in regulating enzyme activities.
