A single-cell guide to retinal development: Cell fate decisions of multipotent retinal progenitors in scRNA-seq

Fion Shiau et al.

Summary: Recent advancements in high throughput single-cell RNA sequencing technology have allowed researchers to simultaneously study the transcriptomic profiles of thousands of individual cells in various species, including the retina. These studies have deepened our understanding of retinal development, progenitor cell competence, cell fate specification, and differentiation, while also pointing out outstanding questions in the field that could soon be addressed with advances in single-cell data generation and analysis.

Summary: The study explores the roles of transcription factors Atoh7, Pou4f2, and Isl1 in retinal development using bulk RNA-seq and single-cell RNA-seq, revealing the emergence of the RGC lineage in the absence of Atoh7 and the defined competence of RPCs for different retinal fates in the transitional state.

Cell-to-Cell Adhesion and Neurogenesis in Human Cortical Development: A Study Comparing 2D Monolayers with 3D Organoid Cultures

Soraya Scuderi et al.

Summary: In this study, transcriptome and cellular phenotypes of telencephalic organoids (ORGs) and monolayers (MONs) derived from induced pluripotent stem cells (iPSCs) were compared. The results revealed increased proliferation and altered cellular characteristics in MONs, suggesting potential differences in signaling pathways and developmental processes when compared to ORGs.

Integrated analysis of multimodal single-cell data

Yuhan Hao et al.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

Initiation of Otx2 expression in the developing mouse retina requires a unique enhancer and either Ascl1 or Neurog2 activity

Michael L. Kaufman et al.

Summary: This study identified a regulatory network controlling Otx2 expression in mouse retina development, with bHLH transcription factors Ascl1 and Neurog2 acting redundantly or compensatorily to activate Otx2. They found that the mechanisms governing Otx2 regulation in the retina are flexible and robust, with redundancy or compensation at both transcriptional and enhancer utilization levels.

Generation of excitatory and inhibitory neurons from common progenitors via Notch signaling in the cerebellum

Tingting Zhang et al.

Summary: This study reveals that individual progenitors in cerebellar development can give rise to both inhibitory and excitatory neuronal lineages. The fate of the progenitors and their daughters is determined by gradients of Notch activity, with varying levels of Notch signaling resulting in retention of progenitor fate, generation of inhibitory neurons, or adoption of the excitatory fate. This Notch-mediated binary cell fate choice mechanism regulates the ratio of excitatory to inhibitory neurons from common progenitors.
