Decoding Cellular Mechanisms for Mechanosensory Discrimination

Summary: Single-cell RNA-sequencing and in vivo functional imaging were used to investigate the molecular and cellular mechanisms of touch detection and encoding in the mammalian mechanosensory neurons. A transcriptomic logic responsible for the sensitivity and selectivity of these neurons was uncovered, with differential tuning of mechanosensory neurons by Piezo2 depending on cell class. This study demonstrates how mechanical stimuli recruit characteristic ensembles of transcriptomically defined neurons, providing rules to help explain touch discrimination.
