An Artemisinin Derivative ART1 Induces Ferroptosis by Targeting the HSD17B4 Protein Essential for Lipid Metabolism and Directly Inducing Lipid Peroxidation

Jingjing Xie et al.

Summary: This study reports that an artemisinin analog, ART1, can induce ferroptosis in a subset of cancer cell lines. The structure-activity relationship analysis reveals that both the endoperoxide moiety and the artemisinin skeleton are required for the anti-tumor activity of ART1. Aided with ART1-based small-molecule tools, the study identifies the HSD17B4 protein as a direct target of ART1, which is crucial for the initiation of ferroptosis.

NIR-Actuated Remote Activation of Ferroptosis in Target Tumor Cells through a Photothermally Responsive Iron-Chelated Biopolymer Nanoplatform

Chencheng Xue et al.

Summary: A polydopamine-based nanoplatform was developed for efficient loading of Fe2+ and beta-lapachone, which could trigger ferroptosis in tumor cells under near-infrared light. The platform could generate mild hyperthermia under NIR irritation, activate cellular heat shock response, and enhance intracellular H2O2 formation, promoting Fe2+-mediated lipid peroxidation and beta-lapachone release.

Polyphenol-cisplatin complexation forming core-shell nanoparticles with improved tumor accumulation and dual-responsive drug release for enhanced cancer chemotherapy

Jiajia Xiang et al.

Summary: In this study, a novel polyphenol-metal coordination method was utilized to fabricate core-shell nanoparticles incorporating CDDP, which were stable in blood circulation but dissociated in tumors to release the drug. Compared to traditional polycarboxylic acid complexation, this strategy achieved higher drug loading contents and faster tumor-targeted drug release effects.

Low dose shikonin and anthracyclines coloaded liposomes induce robust immunogenetic cell death for synergistic chemo-immunotherapy

Jinbo Li et al.

Summary: The study developed a liposome with remote loading capabilities to effectively induce high dosage ICD in vivo. Combining shikonin with anthracyclines in co-loaded liposomes resulted in a synergistic ICD effect and cytotoxicity to tumor cells. Copper-mediated dual-loaded liposomes showed pH and GSH dual-responsive release properties, enhancing antitumor effects at lower drug doses.

Tumor chemical suffocation therapy by dual respiratory inhibitions

Yingying Xu et al.

Summary: The study proposes a novel therapeutic strategy of tumor suffocation by concurrently suppressing glycolysis and mitochondrial oxidative phosphorylation through the co-deliveries of EDTA and rotenone into a glutathione-overexpressed tumor microenvironment. EDTA blocks glycolytic pathway by chelating magnesium ion to inhibit glycolytic enzymes, while rotenone inhibits mitochondrial OXPHOS. This work provides a new approach to treating tumors by inhibiting glucose metabolism, especially by de-functioning glycolytic enzymes.
