New perspectives in oral peptide delivery

Alessandra T. Zizzari et al.

Summary: Peptides, due to their structural diversity, are considered a unique source of innovative active ingredients, but face challenges in development due to their disadvantageous pharmacokinetic properties. Recent efforts have focused on improving the oral bioavailability of peptide drugs, with the latest findings expected to influence future approaches in developing more efficient and safer orally delivered peptides by pharmaceutical companies.

Targeted oral peptide delivery using multi-unit particulates: Drug and permeation enhancer layering approach

Puneet Tyagi et al.

Summary: This work employed layering techniques in a fluidized bed dryer to create a configuration for oral delivery of peptides, which involved coating active peptides, permeation enhancers, and polymers to control release. Formulations were designed to disintegrate at different pH values and trigger release at specific sites in the gastrointestinal tract, with dissolution studies confirming the duration of release. Pharmacokinetic studies in beagle dogs showed that a high disintegration pH was beneficial in enhancing bioavailability.

Once-Weekly Semaglutide in Adults with Overweight or Obesity

John P. H. Wilding et al.

Summary: The study demonstrates that for individuals with obesity, once-weekly use of 2.4 mg semaglutide in combination with lifestyle intervention can lead to sustained and clinically relevant weight loss, with significant improvements in cardiometabolic risk factors and physical functioning reported by participants.

An update on oral drug delivery via intestinal lymphatic transport

Zichen Zhang et al.

Summary: Orally administered drugs must survive the gastrointestinal environment, penetrate the enteric epithelia, and avoid hepatic metabolism before reaching the systemic circulation. Hepatic metabolism is a significant barrier, and alternative pathways like intestinal lymphatic transport are being explored. Main pathways for peroral lymphatic transport include the chylomicron and microfold cell pathways, with increasing research input and applications.
