Overexpressed P75CUX1 promotes EMT in glioma infiltration by activating β-catenin

Anqi Xu et al.

Summary: The study identified a significant increase in the expression of P75CUX1 in glioma, positively correlated with tumor grade, serving as an independent prognostic indicator. P75CUX1 was found to regulate the EMT process mediating glioma infiltration, and promote glioma progression through multiple signaling pathways.

Cut-like homeobox 1 (CUX1) tumor suppressor gene haploinsufficiency induces apoptosis evasion to sustain myeloid leukemia

Emmanuelle Supper et al.

Summary: This study demonstrates that elevated levels of the anti-apoptotic factor CFLAR promotes apoptosis evasion in AML cells haploinsufficient for the CUX1 transcription factor. CUX1 deficiency alleviates CUX1 repression of the CFLAR promoter, driving CFLAR expression and leukemia survival. These findings establish CFLAR as a potential therapeutic target in poor-prognosis leukemias with CUX1 deficiency.

Loss of a 7q gene, CUX1, disrupts epigenetically driven DNA repair and drives therapy-related myeloid neoplasms

Molly K. Imgruet et al.

Summary: Therapy-related myeloid neoplasms (t-MNs) are high-risk late effects in cancer survivors, with poorly understood pathogenesis. This study highlights the critical role of CUX1 in the DNA repair process of hematopoietic stem and progenitor cells (HSPCs), showing that CUX1 deficiency predisposes mice to therapy-related leukemias.
