Toward robust N-glycomics of various tissue samples that may contain glycans with unknown or unexpected structures

Summary: This study developed a combined method to efficiently separate and identify glycan isomers, which involved LC separation of PA-derivatized N-glycans followed by ESI-MS and MS/MS analysis. The method included deducing structures based on elution positions, full MS, and MS/MS data, as well as modifications with sialic acid and permethylation to determine branch sequences. The results confirmed the presence of characteristic branch sequences in N-glycans from chicken colon and the usefulness of the method for comparative glycome analysis in various tissue samples.
