Summary: Phosphatidylethanolamine (PE) is synthesized by membrane-anchored phosphatidylserine decarboxylase (PSD) in bacteria, utilizing the pyruvoyl moiety to form a Schiff base intermediate for decarboxylation. The X-ray crystal structures of PSD from E. coli reveal the mechanism of PE binding, showing extensive hydrophobic interactions with the fatty acyl chains of the phospholipid and the unique topology of the enzyme in a lipid bilayer environment. Mutagenesis analyses suggest that E. coli PSD primarily employs specific residues for auto-cleavage and proenzyme maturation.
