Substrate Tunnel Engineering Aided by X-ray Crystallography and Functional Dynamics Swaps the Function of MIO-Enzymes

Summary: The study reveals that a small opening of the inner loop is sufficient for ligand egress in both eukaryotic aromatic amino acid ammonia-lyases and aminomutases, and the ligand-binding tunnels in these enzymes are analogous. Additionally, engineering of these binding tunnels can convert an (R)-aminomutase to a highly selective (S)-beta-lyase, establishing a novel enzyme function.

Identification and Characterization of an Efficient Phenylalanine Ammonia-Lyase from Photorhabdus luminescens

Summary: A putative aromatic amino acid ammonia-lyase gene called Pl-pal was identified in Photorhabdus luminescens DSM 3368, demonstrating its function as a phenylalanine ammonia-lyase. The enzyme Pl-PAL was able to convert l-phenylalanine and l-tyrosine into trans-cinnamic acid and p-coumaric acid, respectively, with optimal temperature and pH at 40 degrees C and 11.0, providing a promising strain for industrial production of trans-cinnamic acid.
