Reactivation of the pluripotency program precedes formation of the cranial neural crest

Antoine Zalc et al.

Summary: A neuroepithelial precursor population characterized by pluripotency transcription factors is essential for craniofacial development, giving rise to cranial neural crest cells. Through transient reactivation of the pluripotency factor Oct4, cranial neural crest cells expand their developmental potential to differentiate into mesenchymal cells. The open chromatin landscapes of Oct4(+) CNCC precursors resemble those of epiblast stem cells, suggesting priming for mesenchymal programs.

Disruption of RFX family transcription factors causes autism, attention-deficit/hyperactivity disorder, intellectual disability, and dysregulated behavior

Holly K. Harris et al.

Summary: These individuals with de novo or inherited deleterious variants in RFX3, RFX4, and RFX7 share neurobehavioral features including ASD, intellectual disability, and/or ADHD, as well as hypersensitivity to sensory stimuli and sleep problems. The RFX family genes are strongly expressed in developing and adult human brain, with enriched X-box binding motifs and RFX ChIP-seq peaks in the cis-regulatory regions of known ASD risk genes, indicating a likely role of these genes in cases of monogenic intellectual disability, ADHD, and ASD.

Chemical profiling of DNA G-quadruplex-interacting proteins in live cells

Xiaoyun Zhang et al.

Summary: The CMPP strategy allows for the investigation of DNA G-quadruplex (G4) interactions with proteins in native chromatin. This approach enables the identification of G4-interacting proteins in live cells, providing a chemical strategy to study molecular interactions in cellular chromatin.
