Orthogonal translation enables heterologous ribosome engineering in E. coli

Natalie S. Kolber et al.

Summary: The study explores the potential of engineering heterologous ribosomes in living cells, discovering that optimized rRNA processing and supplementation with cognate r-proteins can enhance their function. It also notes reduced subunit exchange with host ribosomes for some heterologous ribosomes. This work provides a general framework for heterologous ribosome engineering and expands the use of heterologous parts in synthetic biology.

Insights into genome recoding from the mechanism of a classic+1-frameshifting tRNA

Howard Gamper et al.

Summary: The study investigates the translation of quadruplet codons by a +1-frameshifting tRNA SufB2, revealing that SufB2 initially uses triplet anticodon-codon pairing in the 0-frame to decode the quadruplet codon before shifting to the +1-frame during tRNA-mRNA translocation. The frameshifting mechanism of SufB2 involves perturbation of a crucial ribosome conformational change, highlighting the potential for engineering specific ribosome conformational changes to enhance genome recoding efficiency.

Sense codon reassignment enables viral resistance and encoded polymer synthesis

Wesley E. Robertson et al.

Summary: By deleting tRNAs in cells through experimental compression and evolution, these cells became completely resistant to viruses and were unable to read the canonical genetic code. By reassigning codons, the cells were able to efficiently synthesize proteins containing different noncanonical amino acids. Additionally, the cells were successfully reprogrammed to translate diverse noncanonical heteropolymers and macrocycles.
