LD-transpeptidases: the great unknown among the peptidoglycan cross-linkers

Summary: The peptidoglycan (PG) cell wall is crucial for bacterial shape and viability, with protective functions due to its mesh-like structure. Penicillin-binding proteins (PBPs) are important targets for antibiotics, while LD-transpeptidases (LDTs) have been less studied but are now recognized for their role in cell wall adaptation to stress, outer membrane stability, and toxin delivery in some bacteria. Further research is needed to fully understand the biological significance of LDTs and their potential as main PG-transpeptidases in certain species.

Only isolates producing multiple septa are able to develop resistance. Defects in PBP1A glycosyltransferase activity and LD-transpeptidase activity play a role in remodeling the cell envelope to support colistin-resistant LOS- A. baumannii.

Essential Gene Analysis in Acinetobacter baumannii by High-Density Transposon Mutagenesis and CRISPR Interference

Summary: The poorly understood bacterium Acinetobacter baumannii poses a serious threat in hospitals due to its high level of resistance to antibiotics. By utilizing dual transposon systems for saturating mutagenesis and developing a CRISPR interference system for efficient gene analysis, essential genes in A. baumannii were identified, including a novel cell division protein and a previously unknown transcription factor. Additionally, it was found that capsule biosynthesis is conditionally essential, with mutations causing toxicity that can be bypassed by blocking early-acting steps or activating stress responses. These findings provide new insights for combating drug-resistant pathogens and understanding their essential biology.
