Multiple cannabinoid signaling cascades powerfully suppress recurrent excitation in the hippocampus

Kyle R. Jensen et al.

Summary: Recurrent excitatory neural networks in the hippocampus involving excitatory mossy cells (MCs) can promote epilepsy. Physiologically relevant activity patterns of MCs induce robust presynaptic long-term potentiation (LTP) of MC-GC transmission. Type-1 cannabinoid receptors (CB1Rs) at MC axon terminals play a crucial role in modulating presynaptic metaplasticity and excitatory transmission onto granule cells (GCs).

Summary: Cannabidiol significantly reduced TSC-associated seizures compared with placebo, with the 25-mg/kg/day dosage having a better safety profile. Both dosages were equally efficacious in reducing seizures, but the smaller dosage led to fewer adverse events.

Chronic cannabidiol (CBD) administration induces anticonvulsant and antiepileptogenic effects in a genetic model of epilepsy

Willian Lazarini-Lopes et al.

Summary: Chronic administration of CBD in a genetic model of epilepsy showed anticonvulsant and antiepileptogenic effects by reducing brainstem and tonic-clonic seizures, preventing limbic recruitment, and suppressing limbic seizures. CBD also prevented chronic neuronal hyperactivity and modulated the expression of CB1R in key brain structures associated with seizures. These results support the chronic use of CBD for epilepsy treatments.

In vivo endocannabinoid dynamics at the timescale of physiological and pathological neural activity

Jordan S. Farrell et al.

Summary: The endocannabinoid 2-arachidonoylglycerol, not anandamide, is found to be dynamically linked to hippocampal neural activity in awake behaving mice. Additionally, seizures significantly amplify the levels of endocannabinoids and drive the synthesis of vasoactive prostaglandins, culminating in stroke-like events. These findings provide new insights into normal and pathological endocannabinoid signaling in vivo.

Summary: Research suggests that current treatments for CAE are not always effective, and modulation of certain cannabinoids and the CB1R receptor may offer more promising pharmacological treatments for absence seizures.
